چکیده
The objectives of DILI WP3 were to qualify one or a set of new biomarkers with respect to: an early or earlier diagnosis of DILI as compared to current diagnostic rules the ability to predict DILI outcome, with particular emphasis on severe DILI/acute liver failure the prognosis and monitoring of progression and regression of DILI the differentiation between patients who incur true drug-induced liver injury from those who recover from the initial injury despite ongoing drug treatment (adaptors)

Drug Induced Liver Injury: Can Biomarkers Assist RUCAM in Causality Assessment?
Drug induced liver injury (DILI) is a potentially serious adverse reaction in a few susceptible individuals under therapy by various drugs. Health care professionals facing DILI are confronted with a wealth of drug-unrelated liver diseases with high incidence and prevalence rates, which can confound the DILI diagnosis. Searching for alternative causes is a key element of RUCAM (Roussel Uclaf Ca...
متن کاملBiomarkers in DILI: One More Step Forward
Despite being relatively rare, drug-induced liver injury (DILI) is a serious condition, both for the individual patient due to the risk of acute liver failure, and for the drug development industry and regulatory agencies due to associations with drug development attritions, black box warnings, and postmarketing withdrawals. A major limitation in DILI diagnosis and prediction is the current lac...
متن کاملElevated levels of circulating CDH5 and FABP1 in association with human drug‐induced liver injury
BACKGROUND & AIMS The occurrence of drug-induced liver injury (DILI) is a major issue in all phases of drug development. To identify novel biomarker candidates associated with DILI, we utilised an affinity proteomics strategy, where antibody suspension bead arrays were applied to profile plasma and serum samples from human DILI cases and controls. METHODS An initial screening was performed us...
متن کاملAssessment of DCEMRI with gadoxetate as a biomarker of drug induced cholestasis
INTRODUCTION One of the major patterns of drug-induced liver injury (DILI) observed in man is cholestatic DILI. The mechanisms are complex and include decreased bile formation, which results in increased bile acid levels in blood [1]. Hepatobiliary transporters mediate uptake and excretion of drugs and endogenous substances. It has been proposed that inhibition of biliary efflux transporters ma...
متن کاملUpdate on Advances in Research on Idiosyncratic Drug-Induced Liver Injury
Drug-induced liver injury (DILI) is a major concern for public health, as well as for drug development in the pharmaceutical industry, since it can cause liver failure and lead to drug withdrawal from the market and black box warnings. Thus, it is important to identify biomarkers for early prediction to increase our understanding of mechanisms underlying DILI that will ultimately aid in the exp...
